Introduction

Degenerative diseases have become a significant health problem in recent years, with an increasing prevalence in the community. One of the primary causes of these diseases is the emergence of hydroxyl radicals in the body, which plays a role in oxidative stress due to an imbalance between free radical production, or what is known as Reactive Oxygen Species (ROS), and antioxidants. When the amount of free radicals exceeds the antioxidant ability to neutralize, the negative effect on cells will appear, including damage by oxidation. This study aims to explore the cytoprotective effect of the Leaves of Building (Coleus Amboinicus Lour) on vero cells induced by hydrogen peroxide (H2O2). The leaves used in this study were taken from Kubuan Village, Tanjung Pura, Langkat, North Sumatra. The use of natural compounds as antioxidants has gained significant attention in recent years due to their potential therapeutic benefits.

Materials and Methods

The leaves of the shape are extracted using multilevel maceration methods with N-HEXANA, ethilatate, and ethanol solvents. Cytoprotective testing is carried out in vitro using the MTT method in various extract concentrations, namely 12.5-200 μg/ml, against vero cells induced by 0.8 mm H2O2. The data obtained were analyzed statistically using ANOVA and continued with the Post Hoc Duncan test using the SPSS version 17 program. The MTT method is a widely used technique for measuring cell viability and is based on the reduction of a tetrazolium salt to formazan by mitochondrial enzymes.

Results

From the results of cell viability analysis, it was found that the treatment with ethyl acetate and ethanol extract (EADBB and EEDBB) increased the percentage of vero cell viability, respectively 74.42 ± 0.28% and 76.08 ± 0.91% at a concentration of 50 μg/ml for EADBB and 100 μg/ml for EEDBB. This shows that the two extracts are more effective than the N-HEKEXANA (ENDBB) extract which do not show significant results. The results suggest that the ethyl acetate and ethanol extracts have a potential cytoprotective effect against vero cells induced by hydrogen peroxide.

Furthermore, testing using the cytometry flow shows that EEADBB and EEDBB can reduce the apoptosis of Vero cells, which increase the number of living cells by 32.14% and 49.59%, while the cell group that is only exposed to H2O2 only shows 10.32% of living cells. This shows a very promising potential from the extract of the Leaves of Building in protecting cells from death due to oxidative induction. The results suggest that the extracts can reduce apoptosis and increase cell viability.

Further analysis using the immunocytochemical method shows that the two extracts (EEADBB and EEDBB) are also significant in reducing the number of ROS in Vero cells induced by H2O2. This is indicated by a reduction in chocolate coloring in cells that are treated with extracts, compared to cells that are only exposed to H2O2. The results suggest that the extracts can reduce ROS levels and protect cells from oxidative stress.

Discussion

Based on the results of this study, it can be concluded that the Leaf-Bangun-Bangun Extract, especially EADBB and EEDBB, show the ability as an effective cytoprotective compound against vero cells induced by hydrogen peroxide. This study opens the potential for the use of these plants as a natural source of antioxidants in therapy for degenerative diseases. The results suggest that the extracts have a potential therapeutic benefit in protecting cells from oxidative stress.
