Introduction

Bacterial wilt in potato plants is a serious challenge for farmers worldwide, caused by the bacterium Ralstonia Syzygii Subsp. Indonesiansis (RSI). Various efforts have been made to control the growth of these bacteria, ranging from land sanitation to the use of bactericides. However, these methods often have limitations, such as environmental risks and reduced effectiveness over time. Background

Methodology

The method used was a complete non-factorial random design with three replications and six treatments:

R4 (RSI 2 with bacteriofage)
R5 (RSI 3 without bacteriophage)
R6 (RSI 3 with bacteriofage)

Results

The results showed that the application of bacteriophage can reduce the number of RSI populations after 24 hours of observation. This is measured through the optical density value (OD) at a wavelength of 600 nm and growth in the NA (Nutrient Agar) media. The best treatment is produced from R6 (RSI 3 isolates with bacteriofage) with a RSI population reaching 1 x 10^8 CFU/ml.

Plaque Testing

In plaque testing, the highest density of BTF1 bacteriofage is obtained in a 10^-6 dilution with a total of 8.65 x 10^7 PFU/ml. This indicates that the bacteriofage has a high infectivity rate and can effectively lyse the RSI bacteria.

Conclusion

This study provides evidence that bacteriofage has a significant potential in suppressing RSI growth in vitro through the cytic infection cycle. The results show that the application of bacteriophage can reduce the number of RSI populations, and the plaque testing results indicate a high infectivity rate of the bacteriofage. Therefore, this study suggests that bacteriofage can be a promising alternative in controlling bacterial wilting diseases in potatoes, potentially replacing the use of chemicals that are more risky.
